چکیده

CD34' precursors in normal human bone marrow (BM) generate large numbers of dendritic cells alongside macrophages and granulocytic precursors when cultured for 12 to 14 days in c-kit ligand, granulocyte-macrophage colonystimulating factor (GM-CSF), and tumor necrosis factor-a (TNF-a). This study reports an intermediate cell type that develops by day 6, and has the potential to differentiate into either macrophages or dendritic cells. When the d6 progeny are depleted of mature macrophages and residual CD34' precursors, a discrete CD14' HLA-DR' population persists in addition to immunostimulatory CD14HLADR'" dendritic cells. Half of the CD14+ HLA-DR' population is in cell cycle (Ki-67'). but colony-forming units (CFUs) are no longer detectable. The cells are c-fms+. but lack myeloperoxidase and nonspecific esterase. They also possess substantial phagocytic and allostimulatory activity. These post-

Hemozoin Enhances Maturation of Murine Bone Marrow Derived Macrophages and Myeloid Dendritic Cells

Background: Falciparum malaria is a severe health burden worldwide. Antigen presenting cells are reported to be affected by erythrocytic stage of the parasite. Malarial hemozoin (HZ), a metabolite of malaria parasite, has adjuvant properties and may play a role in the induction of immune response against the parasite. Objective: To determine the immunological impact of hemozoin on the capacity ...

The role of nanoliposome bilayer composition containing soluble leishmania antigen on maturation and activation of dendritic cells

Objective(s): Dendritic cells (DCs) play a critical role in activation of T cell responses. Induction of type1 T helper (Th1) immune response is essential to generate protective immunity against cutaneous leishmaniasis. The intrinsic tendency of liposomes to have interaction with antigen-presenting cells is the main rationale to utilize liposomes as antigen carriers. In the present study, the e...

Dendritic cells and macrophages can mature independently from a human bone marrow-derived, post-colony-forming unit intermediate.

CD34+ precursors in normal human bone marrow (BM) generate large numbers of dendritic cells alongside macrophages and granulocytic precursors when cultured for 12 to 14 days in c-kit ligand, granulocyte-macrophage colony-stimulating factor (GM-CSF), and tumor necrosis factor-alpha (TNF-alpha). This study reports an intermediate cell type that develops by day 6, and has the potential to differen...

Calcitriol modulates the effects of bone marrow-derived mesenchymal stem cells on macrophage functions

Objective(s):Some evidence showed that calcitriol has an important role in regulating growth and differentiation of mesenchymal stem cells (MSCs). However, the interaction between mesenchymal stem cells and macrophage is not clear yet.  The current study was done to investigate the in vitro effects of calcitriol on the interactions between bone marrow-derived MSCs and rat macrophages. Material...

Optimized Mouse BMDC Isolation and Culture under Endotoxin-Free Conditions

Introduction: Dendritic cells are very important in basic studies and vaccine research, but isolation and culture of these cells face challenges due to their small number in tissues. Since there is no standard method, we addressed some of the factors affecting the efficiency of dendritic cell isolation and culture from BALB/c mouse bone marrow.   Materials & Methods: Bone marrow cells isolated...
